我们计划针对人类纹状体所做的实验，是根据十年前在猴子身上做过的一些实验，希望借此了解人脑视什么为报偿、这个生物过程如何决定人的需求，以及人如何着手满足这项需求。
弗里堡大学的瑞士籍神经科学家舒兹利用猴子做实验，在给予它们各种报偿之时测量纹状体神经元的活动情形。他获得的结果也正是里德丢下松饼的原因。当舒兹在猴子的舌尖滴一小滴（显然很受猴子喜爱的）果汁，猴子的纹状体神经元活动会瞬间暴增。但是当舒兹在滴果汁前提供一条中性线索（如亮起灯泡），激发纹状体神经元活动的不再是果汁而是灯泡——最早预告愉快事物的事件。①这是个惊人的发现，因为灯泡本质上并无好处，也就是说纹状体发出的信号不只是报偿，还有报偿的“预期”。光是预期就能为猴子一以及人类——提供高度的动机。
“你看到舒兹最新的实验结果了吗？”里德问，“看来可以充分证实多巴胺预测误差模式。”

我们啜着咖啡，认真思考这个难题。里德瞪着眼前已喝光的柳橙汁杯子说：“何不用果汁？”“像舒兹的猴子那样？”里德精神为之-振。“对，好极了，我们只要把舒兹的实验复制在人身上就好了。把某种果汁送到扫描仪里的人口中，当然没有问题。”
“我想我们可以架设一些管线，一端放在受试者口中，扫描时把果汁注入他们嘴里。”
里德猛地将最后一块松饼塞入口中，说道：“就这么办。”
就在我们灵光乍现后不久，却发现几乎没有任何资料显示人脑利用哪个部位处理果汁的信息。第一个实验，我们决定在每位参与者的舌头上滴一滴Kool-Aid果汁，但偶尔会以清水代替果汁作为控制条件。实验中主要的操作在于以可预期或不可预期的方式注入果汁和水，前者是固定时间交换注入，后者则是不固定时间与次序地随意注入。如果人脑的反应和舒兹的猴子一样，那么不可预期的注入将是最有价值的报偿，也将最能活化纹状体的活动。
直接测量人脑中的多巴胺是不可能的事——至少在活人身上不可行。虽然可以利用正电子断层扫描（PEf）定位多巴胺受体，以目前的科技却仍无法测量人体释放多巴胺的确实情形。PET法便是对人体注射一种会与脑内特定受体一本例中即为多巴胺受体——结合的放射性追踪剂。圈在受试者头上的侦测器侦测到脑部释放出的放射性物质后，再由电脑判定放射性物质的来源与浓度，因此借由PET可以看到多巴胺受体在脑内不同部位的分布位置与密度——但也仅此而已。通过PET得到的是多巴胺系统的静态快照，而不是多巴胺在脑内时时刻刻活动变化的画面。另外有一种以磁共振成像为基础的间接法是仅次于PET的好方法。我和里德利用一种名为功能性磁共振成像（MRI）的技术，测量出释放多巴胺的脑部区块的血流变化，而且可以看到几秒钟内发生的变化，不像PET需要以分钟或小时为单位。在观察脑部受到各种刺激一例如Kool-Aid果汁——所产生的血流变化后，我们可以根据经验猜测多巴胺可能发生的情形。
里德似乎不太乐意进入MRI仪器内。MRI扫描器是一种非常特殊的医学科技仪器，必须有独立的使用空间。
