Author Description

彭双清，博士，研究员，博士生导师。现为军事医学科学院疾病预防控制所毒理学评价研究中心主任。1983年获学士学位、1986年获硕士学位、1993年获博士学位。1993～1995年在北京大学预防医学博士后流动站进行博士后研究工作。1995～1999年任军事医学科学院毒物药物研究所副研究员，军事毒理学研究室主任。1999～2002年留学美国，在密西根州立大学药理学毒理学系从事神经毒理学研究。2002年任研究员。2004～2007年任军事医学科学院毒物药物研究所新药安全评价研究室主任、国家北京药物安全评价研究中心主任、卫生毒理学教研室主任。多年来一直从事毒理学与药物安全性评价的研究，研究领城为细胞与分子药理毒理，重点研究方向为毒理学替代法。先后主持与参与科研课题20余项，其中包括国家自然科学基金重点课题、国家“863计划”项目、国家科技支撑计划课题、全军医药卫生科研课题等。先后在国内外发表科研论文120余篇，在Mol Pharmacol，JPET和Toxicol&ApplPharmacol等国际知名杂志发表学术论文20多篇。6项科研成果先后获国家、军队科技进步奖。2002年享受国务院政府特殊津贴。2004年获军队杰出人才岗位津贴。多年从事药物毒理学安全性评价和GLP管理工作，主持完成多项药物安全性评价研究。承担国家科技部支持的国家药物安全评价GLP技术平台的建设，负责国家北京药物安全评价研究中心在2005年通过国家GLP认证检查。指导培养博士硕士研究生30多名。任中国毒理学会理事，中国环境诱变剂学会理事，中国毒理学会药物毒理学与安全性评价专业委员会委员、环境与生态毒理学专业委员会常务委员、军事毒理学专业委员会委员，中国环境诱变剂学会风险评价专业委员会委员，中国微生物学会微生物毒素专业委员会委员，是国家食品药品监督管理局新药审评委员、医疗器械审评委员，国家环保部新化学品评审专家，是美国毒理学会、美国神经科学会、中国药理学会等学会的会员，担任《军事医学科学院院刊》、《中国毒理学通讯》、《军事毒理学通讯》、《毒理学杂志》、《生态毒理学报》、《食品安全质量检测技术》等杂志编委。
郝卫东，毒理学教授，博士生导师。1984年毕业于北京医科大学预防医学专业，1987年毕业于北京医科大学研究生院毒理学专业。1990～1991年和1995～1996年在日本冈山大学做特别研究员，1992年在美国加州大学劳伦斯实验室做研究助理。现任北京大学公共卫生学院副院长、学术委员会及教学委员会副主任，毒理学系主任，预防医学实验教学中心主任。兼任国际杂志“Environmen他 Toxicology and pharmacology”副主编，《癌变·畸变·突变》杂志副主编，《中华预防医学杂志》、《卫生研究》等杂志编委，中国环境诱变剂学会秘书长、致突变专业委员会副主任委员，国际环境诱变剂学会执委，亚洲环境诱变剂学会执委，中华预防医学会理事，中国环境科学学会理事、环境与健康分会副主任委员，中国毒理学会理事、生化与分子毒理专业委员会副主任委员，北京市预防医学会毒理学专业委员会副主任委员，全国继续医学教育委员会学科组成员，北京市继续医学教育委员会学科组成员等。近年来，主持或参加了国家自然科学基金项目、国家自然科学基金重大项目、国家“十五”科技攻关项目、“十一五”科技支撑计划重大项目、科技部科研基金项目、北京市自然科学基金重大项目和北京市自然科学基金项目等科研工作。1997年获北京市优秀青年教师奖，1998年获中国环境科学学会首届青年科技奖，1999年获中国环境科学学会优秀科技工作者奖，2007年获北京市教育创新标兵称号。
伍一军，中国科学院研究生院理学博士，美国佐治亚医学院分子医学与遗传学研究所博士后。现任中国科学院动物研究所研究员，中国科学院研究生院兼职教授，博士研究生导师。主持分子毒理学实验室，专业方向为分子毒理学／分子医学，主要研究领域为神经毒理学、环境毒理学以及药物引起的神经疾病。主要研究兴趣在于环境有害化学物质的神经毒性机制；有机磷化合物引发迟发性神经毒性的分子机理及神经病靶脂酶的生理功能；低剂量环境化学物质的致癌机理；溶血卵磷脂及泛蛋白调节在神经毒性发生过程中的作用等。先后主持中国科学院重点项目、国家自然科学基金项目、中国科学院生物特别支持项目、国际合作项目、国家“863计划”项目、中国科学院知识创新工程重大项目（专题）、国家科技支撑计划项目等多项课题。目前，正带领研究团队开展以化学杀虫剂为代表的环境污染物对生物机体的毒性效应机制研究以及食品安全中的关键问题——农药及内分泌干扰物的复合效应研究。已在国内外重要核心学术期刊上发表研究论文60余篇。

第一章毒理学替代法概述
第一节化学物质危险度评价概述
随着科学技术的发展，新化学物质日益增多。目前，全世界已登记的化学品达数千万种，其中常用的化学品就有7万多种，每年还有上千种新的化学品问世。人们在日常生产和生活中通过各种途径接触工农业产品、食品、日用化学品、医用化学品以及饮水和空气中的化学物质。在这些种类繁多的化学物质中，通常只有较少数的化学物质在现有接触水平下对人体可能具有明显的危险性，而接触的大部分化学物质对健康的潜在影响尚未完全清楚。调查研究发现，大量进入人类生产和生活环境中的外源化学物尚缺乏基本毒理学资料（急性和慢性毒性、发育/生殖毒性、遗传毒性/致突变性、生态毒性、环境影响等），这对人类正确评价这些化学物质的安全性带来了困难，对人类和环境构成了潜在的危害。随着化学物质的增多，时有潜在危害的存在以及中毒事故的发生，对外源化学物的危险度评价显得尤为重要和紧迫，并促使世界各国制定相应的法规，要求对有毒化学物质进行危险度评价，以适应相关管理部门处理灾情、立法、决策和制定卫生标准的需要。
Introduction

毒理学是研究人类生产实践和生活中的各种化学性、物理性及生物性有害因素对生物体，特别是对人体产生危害与毒性作用机制的科学。其主要任务是评价外源性化学物质对接触者可能造成的健康危害，最终为控制化学物质的危害、加强化学物质的管理提供依据。通过对外源性化学物的危害评价和提出对外源性化学物的管理措施，毒理学在保障人类健康、维护生态平衡、改善环境和促进国民经济持续发展等方面发挥着重要的作用。随着科学技术的发展，新化学物质日益增多，传统的毒理学评价方法面临着新的挑战。近年来，随着实验动物使用3R原则（Reduction一减少；Refinement一优化；Replacement一替代）的倡导与实施以及生物医学研究模式的转变，整体动物实验面临严峻挑战，替代动物实验的体外模型研究已成为毒理学发展的重要方向。替代法它包含了3R原则的全部内容，毒理学替代法就是3R原则在毒理学研究领域的应用，它不仅是动物权益保护的需要，也是科学进步、社会经济发展的需要。无论是从科学角度还是从经济角度考虑，毒理学替代法对外源性化学物的危害评价及管理均具有重要的意义，它可以减少体内实验影响因素、减少动物使用、缩短实验周期以及降低实验成本等多种优越性。近年来，国外毒理学替代法研究发展十分迅速，体外替代试验已经涵盖一般毒性、遗传毒性、器官毒性等多种毒性终点，研究手段也从一般的细胞、组织培养延伸到基因组学、蛋白质组学与代谢组学，以及计算机模拟辅助评价系统。毒理学替代法已广泛应用于毒理学研究各个领域，欧盟和美国等发达国家已将毒理学替代法纳入法规管理范围。目前，许多毒理学替代法已通过有关权威机构的验证并被欧盟、美国和经济与合作发展组织（OECI））等推广应用。我国关于化妆品、化学品、食品接触材料以及药品和生物制品等化学物质的安全性评价检测方法，基本上仍使用整体实验动物，毒理学替代法的研究和应用相对滞后，尚没有专门的研究机构和验证体系。虽有实验室和研究人员在这方面进行了有益的尝试和积极的探索，但缺乏工作的系统性和严格的验证程序，只能说处于研究探索阶段。到目前为止，国内尚没有一本系统介绍毒理学替代法的专著。在国家科技支撑计划食品安全专项课题的资助下，近年来我们积极开展了毒理学替代法的研究工作。结合国内外研究发展趋势，我们以国际毒理学替代法权威机构，如欧洲替代方法验证中心（EcvAM）和OEcD验证认可的各种毒理学。
