Author Description

何韶衡，1957年出生，四川三台县人，博士，教育部“长江学者”特聘教授。曾在英国、美国留学12年，获英国皇家学会奖励及英国南安普顿大学超聘一级工资的奖励。1 999年在世界著名的英国南安普顿大学呼吸细胞与分子生物学分院任高级研究员，博士生导师，是分院领导中唯一的亚裔成员。现任南京医科大学第一附属医院临床实验研究中心主任，变态反应研究所所长，江苏省医学领军人才，中国协和医科大学客座教授。主要从事过敏性疾病发病机制的研究及过敏性疾病诊治手段的研发工作，是国际知名专家。2009年入选Whor‘s Who in the World。
共发表论文270余篇，其中60篇被SCI收录，在国内外共主持省级重点以上科研课题20余项，获国家发明专利3项。2005年，获广东省科技进步一等奖。排名第一。

Book Abstract

上皮细胞细胞之间以及上皮细胞与成纤维细胞之间连接紧密，上皮细胞所用的消化酶的浓度与消化时间不易掌握。若消化酶的浓度与消化时间控制不好，使消化不到位，则分离下来的细胞数量较少；消化过度则可能将黏膜层的成纤维细胞连带消化下来，同时可能破坏已消化下来的细胞膜的完整性，影响已分离的上皮细胞的质量，不能很好地贴壁、增殖与应用。
使用酶消化联合机械刷洗法分离培养，不仅简化操作过程，减少污染概率，也使细胞在酶的消化作用下易于脱落，稍加刷洗即可得。采用链酶蛋白酶消化时，细胞易于分散脱落，且对细胞损伤相对较小，结合细胞刷刷洗，不仅可大量回收基底层细胞，且提高了上皮细胞的收获量与存活率。秦晓群等分别比较了酶消化法、机械刷洗以及消化后刷洗3种方法分离家兔气管一支气管上皮细胞，结果表明消化后刷洗方法可得到的细胞数量与存活率高于前两种方法。
利用酶消化联合机械刷洗法获得的上皮细胞纯度与成活率高，收获的细胞数量较多，避免了单纯消化法的细胞得率低、纯度低及存活率低的缺点，尤其是单纯刷（刮）洗方法难免会将成纤维细胞一起刷洗下来。成纤维细胞与上皮细胞共存状态下会造成上皮细胞生长抑制。
经改良的无血清完全F12培养基更适合上皮细胞培养，无血清培养可提高培养上皮细胞的纯度，促进细胞融合，细胞生长状态良好。

过敏反应学在世界上已有100多年的历史，随着人们对过敏性疾病认识的不断加深，尤其是近二三十年来分子免疫学、细胞生物学、生物信息学、基因组学和蛋白质组学等学科的迅猛发展，使过敏反应学领域的研究水平不断提升，新的突破不断出现，研究内容更是日新月异，成为医学乃至生命科学研究工作一个极为活跃的领域。
过敏反应学的迅猛发展对研究和诊断手段提出了更高的要求，该书从细胞生物学技术在过敏反应学中的应用、生物化学与分子生物学技术在过敏反应学中的应用、蛋白质组学与生物信息学在过敏反应学中的应用、抗体技术在过敏反应学中的应用、过敏反应相关的动物实验模型、临床相关的过敏反应学实验技术以及常见过敏原的收集与鉴定人手，系统地阐述了过敏反应学的实验室研究和诊断的方法，是对近20年来国内外相关领域进展的一次总结，为从事过敏反应学相关工作的基础研究者、临床工作者以及研究生等人员提供了一部重要的参考书。
