Book Abstract

分配层析中，通常采用一种多孔性固体支持物（如滤纸、硅胶、纤维素粉、淀粉等）附着的一种溶剂作为固定相，另一种与固定相溶剂互不相溶的溶剂可以沿固定相流动，称为流动相。以滤纸作为支持物的分配层析，称为纸层析。纸层析固定相指滤纸结合的水，流动相则为不溶性的非极性溶剂（展开剂）。展开时，非极性展开剂沿滤纸流经样品点，样品中各物质在两相之间不断地进行分配，Kd大的物质留在固定相中相对多，随流动相移动得较慢；反之亦然。
3.应用
纸层析法的定性和定量方法与薄层吸附层析类似。
纸层析法在临床生化检验中应用广泛，可用于血液氨基酸、类固醇激素、糖蛋白、儿茶酚胺代谢产物等的分析，以及安眠药、抗生素等药物在体内代谢情况的检测。
三、离子交换层析（ion—exchange chromatography）
1.原理
离子交换层析是利用离子交换剂对样品离子的亲和力不同，借以分离混合物中各种离子的一种方法。根据离子交换剂所连接的交换基团不同，又分为阳离子交换层析和阴离子交换层析。
离子交换层析的固定相是带有大量电荷的离子交换剂，流动相是具有一定pH值和离子强度的电解质溶液。当混合物溶液中带有与离子交换剂相反电荷的溶质流经离子交换剂时，被离子交换剂选择性吸附。然后提高流动相离子强度或改变其pH值，被吸附的溶质可被置换而洗脱下来，结合弱的分子被先洗脱下来，结合强的被后洗脱下来，从而达到分离混合物中各种带电荷溶质的目的。
如图示3—2所示，自身带正电荷的阴离子交换剂必须吸附带负电荷的离子以维持电中性。当样品离子流经阴离子交换剂时，其中的阴离子被吸附。样品离子带负电荷越多，与阴离子交换剂的亲和力越大，结合越紧密，洗脱过程中被洗出也越慢。相反带负电荷越少的离子则越先被洗脱出来。
