目前，医学细胞生物学已为国内多数院校五年制本科生基础阶段培养的必修课。在多年的教学实践中，编者感到同学们在经过大学一年级学习之后，对《人体解剖学》、《组织胚胎学》等需要记忆的课程逐步适应，而对《医学细胞生物学》等在二年级开设的主修课程应付起来却略显茫然。为此，编者编写了《医学细胞生物学难点解析与应试指南》一书，一来解决同学们学习中可能遇到的问题；二来帮助同学们归纳整理学习要点，紧紧抓住医学细胞生物学这门课程的精髓所在。

2.微丝组装受哪些因素影响？
答：微丝的装配受肌动蛋白临界浓度的影响。在正常的体外条件下，单体的临界浓度（critical concentration，Cc）是0.1μM。高于该值，G-肌动蛋白倾向于聚合，低于该值，F-肌动蛋白将会解聚。
在肌动蛋白纤维的装配过程中，除了受G-肌动蛋白临界浓度的影响，还受一些离子浓度的影响。如向G一肌动蛋白溶液中添加Mg抖、K+、Na十，可诱导G一肌动蛋白聚合成F-肌动蛋白。该过程是可逆的，当这些离子的浓度较低时，F-肌动蛋白趋于去聚合，而在Mg2+和高浓度K+或Na+的溶液诱导下，G-肌动蛋白则装配成纤维状肌动蛋白。
3.中等纤维在细胞中有哪些功能？
答：近年来，采用转基因（gene transfer）和基因删除（gene deletion）小鼠研究中等纤维蛋白的功能，证实中等纤维在细胞的生命过程中具有多方面的功能并且起相当重要的作用。归纳其主要功能如下：①具有支持作用；②在细胞内的运输作甩；③具有信息传递作用；④在相邻细胞、细胞与基膜之间形成连接结构；⑤维持细胞核膜的稳定。
4.简述纤毛、鞭毛的运动机制。
答：纤毛和鞭毛都是某些细胞表面的特化结构，具有运动功能。纤毛和鞭毛都含有一个规则排列的由微管相互连接形成的骨架，称为轴丝（axoneme）。纤毛和鞭毛的运动由轴丝中微管动力臂引起微管的滑动所致，即“微管滑动模型（sliding-microtubule model）”其主要内容是：纤毛和鞭毛的动力蛋白头部与相邻二联管的B微管接触，促进同动力蛋白结合的ATP水解，并释放ADP和Pi；由于ATP水解，改变了A微管动力蛋白头部的构象，促使头部朝向相邻二联管的正极滑动，使相邻二联管之间产生弯曲力；新的ATP结合，促使动力蛋白头部与相邻B微管脱离；ATP水解，使动力蛋白头部的角度复原；带有水解产物的动力蛋白头部与相邻二联管的B微管上的另一位点结合，开始下一个循环。
5.简述肌肉收缩的兴奋一收缩耦联过程。
答：兴奋一收缩耦联（excitation-contraction coupling）是指肌肉细胞接受神经末梢的冲动信号产生肌节缩短的过程，包括①动作电位的产生，来自脊髓运动神经冲动经轴突首先传到神经肌肉接点一运动终板，使肌肉细胞膜去极化，再经T小管（transverse tubule）传至包裹在肌原纤维周边的肌浆网（sarcoplasmic retictllum，SR）。②Ca2+的释放，SR膜蛋白中Ca2+转运蛋白占80％，SR去极化使膜Ca2+通道迅速开放，大量的Ca2+释放到肌浆中，浓度可达5×10-6M。③原肌球蛋白移位，Ca2+与TnC结合，引起后者构象变化，TnC与TnI、TnT的结合力增强，TnI抑制肌动蛋白结合力减弱，肌动蛋白因此摆脱TnI的束缚，变成应力状态；同时，TnT使原肌球蛋白移动到肌动蛋白双螺旋沟深处，消除肌动蛋白与肌球蛋白结合的障碍，驱使肌动蛋白与肌球蛋白之间滑动的开始。
