《男性生殖基础与实验室研究》共8章，前5章着重介绍了男性生殖的基本理论知识，包括男性生殖系统的结构与精子发生、精子发生的调控、精子的成熟与获能、精子的受精功能以及生精细胞的凋亡。从结构和功能方面对男性正常生殖的发生机制进行了简要概述。第六、七、八章为实验方法学部分，主要阐述了目前细胞生物学、分子生物学和基因功能分析技术常用实验方法的原理及其在男性生殖基础研究中的应用，这部分是以笔者实验室工作为基础，对其中的某些方法进行了较为详细的介绍，如细胞的分离纯化、免疫组织化学分析技术、免疫荧光技术、核酸提取及电泳、PCR技术、分子杂交和芯片技术、重组DNA技术、RNA干扰技术和基因敲除技术等。书中附有大量图片，可帮助读者加深对相关内容的理解。《男性生殖基础与实验室研究》既涵盖了男性生殖主要的基础理论知识，又阐述了常用的实验室技术方法，对从事人类生殖基础研究的科研工作者具有参考价值。
Author Description

郭睿，女，1972年3月生人，理学博士，副教授，硕士研究生导师。1994年毕业于山西医学院儿科学专业，1997年获山西医科大学免疫学专业硕士学位，200l～2004年攻读中国医学科学院／中国协和医科大学基础医学研究所细胞生物学专业博士并获博士学位。2005年留学欧洲生命科学最高学府——瑞典Karolinska Institutet进行博士后研究。自1997年开始从事生物化学与分子生物学专业的科研及教学工作，先后承担了博士研究生、七年制硕士研究生、本科生和专科生的专业课程讲授，教学效果突出，于2005年作为主讲教师之一荣获由山西省教育厅授予的省级精品课程称号。进行科研工作15年来，先后参与和主持多项科研工作，主要从事分子发育生物学和肿瘤相关基因分子作用机制的研究。目前主持国家自然科学基金项目1项，省级项目3项，获2008年度山西省高等学校优秀青年学术带头人支持计划1项；共发表科研论文23篇，其中SCI收录7篇，国家级核心期刊9篇；参与编写生物化学教材3部。

第二章 精子发生的调控
精子发生是一个复杂而有规律的周期性同步发育过程，受许多特异分子及细胞间作用的严格调节，包括自身染色体结构的变化及一系列特定基因的程序性表达。这些内在基因水平的变化又受许多外部因素如激素、环境因素及旁分泌细胞因子的影响。雄性哺乳类个体大致从内源性、相互性、外源性三个层次对这一过程的基因表达进行调控。
生精细胞内源性机制调控精子发生中的关键事件，如减数分裂、精子变形等特异性活动。该机制调控不同的基因按严格的时间、空间特定表达，以保证精子发生能正常进行。以Sertoli细胞为中心的细胞间相互调节机制在保证精子的周期性同步发育上具有举足轻重的作用。每一个Sertoli细胞都同时与不同发育阶段的3～4层生精细胞紧密接触，还与其他支持细胞以及Leydi9细胞、曲精小管周围细胞发生相互作用，是最适于整合、协调各种信号的中心枢纽。从整体意义上讲，相互性调控机制又受控于以睾酮、黄体生成素（Iuteinizing hormone，LH）和促卵泡激素（follicle stimulatinghormone，FSH）为主的外源性机制的控制，Leydi9和Sertoli细胞表面分别存在LH和FSH受体，LH可刺激Leydi9细胞分泌睾酮，弥散进入曲细精管，在睾酮和FSH的联合刺激下，Sertoli细胞可分泌包括雄激素结合蛋白在内的多种生精细胞分化所需的多肽等物质，从而影响精子发生。因此，Sertoli细胞是精子发生和睾酮、FSH调控的主要靶点，而后二者又调控着精子的发生、分化以至成熟。也就是说，机体内分泌系统通过直接影响睾丸内体细胞的基因表达，来调节相互性作用机制赖以生存的周边环境，进而影响生精细胞的基因表达。
