Book Abstract

消化系统：每天通过消化道和消化腺吸收或分泌的液体多达9L，而且至少有8种水通道蛋白（水通道蛋白一1、—3、一4、一5、一8、一9、一10、—12）在消化道上皮表达。水通道蛋白一5敲除小鼠唾液分泌减少。水通道蛋白一4存在于近端结肠上皮细胞基底膜，水通道蛋白.4敲除小鼠盲肠粪便中的水分与野生型相比无差别，但排泄的粪便中的水分含量明显增大，在体结肠灌流实验证明，水通道蛋白-4敲除小鼠跨膜结肠上皮水的通透性明显降低，提示水通道蛋白.4参与结肠液体的吸收。水通道蛋白一1缺乏引起饮食性脂肪处理缺陷，水通道蛋白.1缺失小鼠脂肪吸收障碍，脂肪泻，这可能与肝胆汁缺乏、胰液缺乏及小肠内脂肪吸收障碍有关。近年来研究表明，肝硬化小鼠腹膜水通道蛋白-蛋白及其基因表达下调，提示水通道蛋白一1可能在腹水形成过程中起重要作用。（4）神经系统：水通道蛋白广泛分布于中枢神经系统。在生理条件下，脑内水通道蛋白主要参与血脑屏障的构建、脑脊液的形成等过程，也有观点认为，水通道蛋白可能参与脑能量代谢。水通道蛋白一3、水通道蛋白一5、水通道蛋白-8 mRNA存在于神经元内；水通道蛋白—3、水通道蛋白一4、水通道蛋白一5、水通道蛋白-8和水通道蛋白一9 mRNA则存在于星形胶质细胞。有研究证实，在脑导水管内皮细胞中也存在水通道蛋白1mRNA。目前对于脑内水通道蛋白的研究多集中在水通道蛋白一4这一亚型上。自水通道蛋白发现以来，对其结构、表达、调节及相关功能的研究已取得阶段性进展，细胞膜水通道蛋白功能紊乱引起的相关疾病逐渐受到人们重视。对水通道蛋白的深入研究可以帮助阐明某些水通道蛋白代谢疾病的发病机制并研制相应的药物。如水通道蛋白抑制剂在利尿、抑制腺体分泌、治疗脑水肿、高血压和青光眼等方面具有广泛应用前景。水通道蛋白的基因治疗研究还有望为口干症、干皮病及尿崩症提供新的治疗方向。此外，大量研究证实，肿瘤的生长与转移与水通道蛋白功能异常有密切关系，因此，水通道蛋白也可能作为肿瘤药物治疗的新靶点。3低氧条件下水通道蛋白的改变脑缺血缺氧可由多种脑损伤引起，如脑出血后血肿压迫周围脑组织，导致周围脑血流量减少，从而形成缺血性脑水肿。有学者通过建立大鼠大脑中动脉栓塞脑缺血后水肿模型，发现与野生型鼠相比，水通道蛋白一4敲除大鼠的星形胶质细胞足突的膨胀程度较轻。持续中动脉阻塞缺血后24 h测定发现，大脑半球肿胀率下降35％，神经病理改变明显减轻，说明水通道蛋白一4在缺血后继发性脑水肿中发挥重要作用。在缺血后第1天，栓塞周围皮质的水通道蛋白一4基因表达轻微上调，第3天达到高峰，第7天仍处于较高水平，与磁共振监测的脑水肿变化相一致。水通道蛋白一4能够促进血管源性脑水肿时过多水的再吸收，从而减轻血管源性脑水肿，提示水通道蛋白一4的激活和（或）上调有可能成为血管源性脑水肿的一种新的治疗手段。水通道蛋白一4在大脑中动脉闭塞模型鼠脑的表达研究发现，水通道蛋白一4在脑梗死灶周围的基因表达增加主要出现在水肿形成的较晚期，尤以梗死灶周围明显，且与脑水肿的程度呈正相关，推测脑缺血后星形胶质细胞中水通道蛋白一4表达增加使得水快速转运而形成细胞内水肿。
